01 / PERSPECTIVE

The signal: AI is entering validation work

The ISPE programme includes AI-enabled computerized system validation and AI-enabled quality systems. These are useful prompts to examine where automation helps a validation team and where it can create convincing but unsupported records. A generated requirement, test or report is a proposed artifact until its basis and execution have been evaluated. [1]

FDA's February 2026 final CSA guidance concerns software used in medical-device production and quality management systems. It should not be presented as a blanket replacement for drug GMP validation. A team needs to establish the applicable framework and intended use for its own system before borrowing an assurance approach. [2]

02 / PERSPECTIVE

Assurance belongs to the quality system and the operating team

Pharma 4.0 combines digital capability with the pharmaceutical quality system, controlled processes and organizational accountability. An AI-assisted validation workflow therefore needs a system owner, qualified reviewers, change responsibilities and a retained explanation of what was actually tested. More generated documentation is not itself a stronger state of control. [4]

Bring QA, the subject-system owner, scientific users and implementation staff into the same risk discussion. Separate the assurance of the drafting assistant from assurance of the system being evaluated. Give reviewers the authority and capacity to reject a convincing test that cannot detect the relevant failure, and preserve that learning in the next revision.

03 / PERSPECTIVE

Separate the subject system from the assistant

Imagine a hypothetical team using AI to draft tests for a laboratory data interface. The interface is the subject system. The drafting assistant is another component with its own use, inputs and limitations. Tests for the interface do not automatically demonstrate that the assistant consistently generates appropriate tests for every interface.

Define the assistant's job narrowly: propose candidate tests from approved requirements and risk analysis for qualified review. Preserve the source requirement and the rationale behind each test. The reviewer must be able to identify missing coverage and remove irrelevant cases. This creates a traceable proposal workflow rather than an automated declaration that the system is validated.

04 / PERSPECTIVE

Follow the requirement through three evidence questions

First, does the subject system perform the required function? Second, does the assistant produce suitable evidence for that function without dropping requirements, inventing observations or selecting an inapplicable source? Third, if a scientific calculation contributes to the result, is its method and applicability justified for the decision?

These questions are connected, but passing one does not answer the others. A well-tested application does not make every generated test correct. A numerically reproducible calculation does not establish that the underlying model represents the process adequately.

Choose a material requirement and follow it through the source, proposed test, actual observation, calculation where relevant, issue handling and qualified conclusion. Include a deliberately omitted requirement or conflicting source. The reviewer should identify the gap and prevent an unsupported completion claim. Scale the evidence to the intended use and consequence of failure, with the applicable regulatory scope established separately.

05 / PERSPECTIVE

Work backward from the failure that matters

For the interface, an important failure might be assigning an analytical result to the wrong batch. Start with the consequence and the conditions that could produce it. Then define a challenge that distinguishes correct behavior from a superficially successful import. A test that only confirms the screen displays a number is weak evidence for record identity.

Our suggested risk discussion covers data identity, calculations, permission, exception handling and the receiving decision. It should involve the users who recognize meaningful failures. The amount and type of evidence should follow the assessed use and risk, with a clear explanation for what was tested and what remains outside the scope. [3]

06 / PERSPECTIVE

Generated tests need adversarial review

Give the assistant an ambiguous requirement and ask a reviewer whether the proposed test can actually detect the relevant failure. Include repeated identifiers, unusual units, delayed records and a partially failed transfer. The point is not to create the longest test pack; it is to evaluate the boundaries that matter for the function.

Keep generated expectations separate from observed results. A proposed expected outcome must not appear as if it were an executed result. Execution records should identify the system version, inputs, actual behavior and disposition of anomalies. If an assistant summarizes those records, the summary should remain traceable to the original execution evidence.

07 / PERSPECTIVE

Evaluate the drafting workflow as a workflow

Count accepted useful tests, missed critical scenarios, reviewer corrections and the time needed to reach an approved test set. An increase in document volume can conceal a decline in useful coverage. A reviewer who spends hours removing repetitive cases has absorbed the work that the automation was meant to save.

Retain difficult examples in a regression set for the assistant. The purpose is to detect whether a changed model, instruction or source representation weakens the drafting task. Success on that set supports a bounded internal conclusion about the assistant; it is not evidence that all future generated validation packages are acceptable without review.

08 / PERSPECTIVE

Manage changes that affect the evidence path

A model update, a new retrieval source or a changed requirement parser can affect the proposed tests. Record the changed component, the functions it influences and the evidence supporting continued use. Different changes may justify different levels of reassessment. The workflow should make that judgment explicit rather than treating every upstream update as invisible maintenance.

Also plan recovery. If a new version generates incomplete coverage, identify the affected outputs and preserve the previous configuration where appropriate. Decide who can pause the assistant and how ongoing validation work continues. Operational continuity is part of useful assurance, especially when the tool becomes embedded in a team's planning process.

09 / PERSPECTIVE

BioMedAna: evaluate the computational layer as well as the generated text

BioMedAna combines agents with implemented numerical services: engineering calculations, chemical reaction simulations and mechanistic/data-driven hybrid models. Assurance should therefore ask more than whether the narrative looks correct. It should inspect input units, calculation versions, supported model ranges, failure behavior and the evidence connecting an output to its intended decision.

Use analytical benchmarks and unit or conservation checks for the relevant numerical function, then evaluate scientific applicability with suitable process evidence. Review the orchestration and the receiving human workflow separately. A software benchmark cannot establish that a model is adequate for every process, and a qualified model does not validate an unsupported sentence generated about its result.

QUICK ANSWERS

The questions behind the headline

Does CSA replace drug GMP validation?

The cited FDA guidance has a medical-device production and quality-system scope. Applicability must be determined for the actual system and regulatory context.

Can AI draft a validation protocol?

It can propose content for a bounded reviewed task. Draft quality, scientific coverage and execution evidence still need assessment.

What is a good early acceptance test?

Ask whether the workflow detects a wrong-batch assignment and preserves the observed failure and reviewer disposition.
